Cigarette smoking is estimated to lead to 4.9 million premature deaths per year worldwide. This is predicted to rise to 10 million by 2020. In western countries it kills half of all smokers who fail to stop. The prevailing model for tobacco addiction is that nicotine from cigarettes rewards smoking and punishes abstinence, tapping into a motivational system of operant conditioning that requires no conscious awareness. However, there are also accounts which involve cognitive biases and the effect of nicotine on impulse control. The brain pathways involved have been studied extensively, but the role of different nicotine receptor subtypes and other neurotransmitter systems is still subject to debate. In western countries, cigarette smoking as an adult has a heritability of 30-50% and candidate genes have been identified that may contribute in part to addiction susceptibility. Many socio-cultural correlates of cigarette smoking have been established, but a comprehensive model that accounts for these and links them with the psychobiological aspects of nicotine addiction has not been forthcoming. Structured behavioural support programmes aid cessation attempts, as do a number of pharmacotherapies, most notably nicotine replacement treatments and bupropion, but the underlying mechanisms are unclear. This book deals with the problems involved in understanding and treating nicotine and tobacco addiction. Topics covered include the nature of the worldwide health problem posed by cigarette smoking, the psychodynamics of cigarette addiction, and the basic pharmacology and biochemistry of nicotine and its effect on the brain. Further chapters analyse the genetic basis of susceptibility to nicotine addiction. Finally, the contributors address approaches to therapy. A continuing theme in the discussions throughout the book is how best to treat nicotine addiction, given that many smokers would like to stop smoking but are unable to do so because of their addiction. Understanding Nicotine and Tobacco Addiction

This book features contributions from experts in cell biology, genetics, neurobiology, immunology and structural biology. The unifying element is that they all study processes of cell shape change and motility. Several key questions in this field of research are discussed: What are the organising principles behind cell shape change? Are there ‘master switches’ present in every cell type? How are extracellular signals interpreted by the cell in order to activate intracellular mechanisms? Signalling Networks in Cell Shape and Motility

Generation and Effector Functions of Regulatory Lymphocytes

Over recent decades vast amounts of biological data have been accumulated. However, it is becoming increasingly difficult to apply traditional theoretical methods to the formulation of coherent pictures of cell and organ function because it is no longer possible for a human theorist to integrate all of the available information. Instead, computer technologies must now be used to perform this integration. This book brings together contributions from many different fields to summarize the current status of computer-assisted modelling of biological processes. The initial chapters deal with fundamental developments in hardware, software and mathematics that underlie current approaches to biological modelling. Next, different approaches to collating data on gene structure and function are presented. These databases form a vital resource for any investigator trying to construct an integrated picture of particular biological systems. Cell signalling systems form a particularly complicated aspect of all cellular function and are important both in the understanding of basic cellular processes and in selecting targets for drugs. The Limits of Reductionism in Biology

Dietary supplements can contain a wide variety of ingredients, either singly or in combination, including nutrients, botanicals and 'bioactive components' commonly found in foods. They are marketed and used by consumers for a range of reasons: to enhance «well-being», as traditional medicines, for health promotion or disease risk reduction, and as alternatives or complements to conventional drug therapies. On a global basis, the dietary supplement industry has enjoyed rapid growth, becoming a multi-billion dollar enterprise over the last 10 years. Anaphylaxis is an immediate-type allergic reaction involving the whole organism. It is the most life-threatening allergic condition. Although there are few exact epidemiological data regarding prevalence, estimates regarding insect sting anaphylaxis range from 1-3% in the general population, but much higher values have been reported by some authors for food and drug-induced anaphylaxis. Anaphylaxis is the main acute killer of allergic individuals. Although anaphylaxis was discovered at the beginning of the 20th century, there are still many unresolved issues. These include non-IgE-mediated anaphylactoid reactions, non-immunologically mediated anaphylactoid (pseudo-allergic) reactions, pathophysiological events at the microcirculatory level, appropriate therapy for the acute reaction, strategies for prevention, public education about the problem and new approaches to prevention and therapy at the IgE level. All these subjects are discussed in this book. Since anaphylaxis occurs acutely and is unforeseen, it is very difficult to organize controlled studies regarding therapy and prevention. Polyfunctional Cytokines

'Homology' as a concept became increasingly elusive during the course of the 20th century. The central debates and controversies concern both fundamental definitions and the nature of the criteria by which homology is judged. Sensory Guidance of Movement Chairman: Mitchell Glickstein, 1998 In the past few years there has been an increasing recognition of the multiplicity of sensory and motor areas of the cerebral cortex. However, still relatively little is known about the way in which sensory areas are functionally linked to motor areas. In order to elucidate the underlying mechanisms of this major clinical condition, this symposium brought together neuroscientists working on the basic biology of the bladder and bowel and clinicians dealing with the various manifestations of urinary and fecal incontinence. The Childhood Environment and Adult Disease

Environmental statistics is a rapidly growing discipline that is important not only as a division of professional and academic statistics, but also in the accumulation of data on environmental effects and in the formulation of environmental policy by government agencies. This book features contributions from policy makers, scientists actively involved in collection of data, and professional, academic and government statisticians. These explore the interfaces between the different areas of application of environmental statistics and consider the future applications of methods arising from this discipline. The potential lipotoxic effect of accumulation of fatty acids in non-adipose tissues is thought to be a major component in the development of insulin resistance. Chronic exposure to high concentrations of free fatty acids in the blood affects pancreatic β cell function, insulin secretion and lipid synthesis in the liver, and storage in adipose tissue. Fatty Acid and Lipotoxicity in Obesity and Diabetes

This book explores existing and potential strategies for using the genome sequences of human, mouse, other vertebrates and human pathogens to solve key problems in the treatment of immunological diseases and chronic infections. The term «unconventional virus» refers to virus-like agents that differ from conventional viruses in significant respects–resistance to agents that inactivate normal viruses, for example. The Origins and Development of High Ability

Ciba Foundation Symposium 123 Antidepressants and Receptor Function Chairman: Dennis Murphy, 1986 Depression is a common and often debilitating affective disorder. Attempts to develop effective antidepressants have a long history, but many questions remain about the mechanisms of action of such treatments and about the aetiology and pathophysiology of depression itself. Early observations centred attention on central monoamine systems, and animal studies suggested that changes in beta-adrenoceptor responsiveness were a common effect of antidepressant therapies. More recent research has encompassed many different central and peripheral receptors, time-dependent adaptational events at synapses, and the functional significance of changes in neurotransmitter systems in both humans and experimental animals. Such pharmacological studies aimed ultimately at elucidating the neurochemical basis of depression and of promoting new therapeutic approaches, provide the focus of this symposium volume. Many different methods of investigating the links between monoamine systems, depression and antidepressant treatments are described. Recent studies of receptors and of monoamine uptake sites in the brain and the periphery (e.g. in platelets and fibroblasts) are reviewed, with emphasis on alpha and beta adrenoceptors, [3H]imipramine-binding sites and serotonin receptors. The results of monitoring amine metabolites in cerebrospinal fluid and of measuring neuroendocrine, physiological and behavioural responses to pharmacological challenge are presented, providing information on monoaminergic function in depressed patients and experimental animals before, during and after treatment with antidepressant drugs or electroconvulsive shock. Genetic influences on receptor density are also discussed, as is the relevance to human depressive illness of animal models, including stress-induced behavioural depression in rats and responses to social stressors in rhesus monkeys. This book should be of interest to neuropharmacologists, psychopharmacologists, clinical pharmacologists, behavioural scientists, psychiatrists and neuroscientists.

Antidepressants and Receptor Function

Genetics of Autoimmunity

Parkinson's disease, Alzheimer's disease, and motor neuron disease share a significant common feature: selective death of neurons in restricted regions of the brain. This international symposium, held by the Ciba Foundation in 1986, is the first to bring together neurophysiologists working on neuronal death and neuropathologists dealing with human degenerative brain disease. Participants describe the causes and sequence of events leading to neuronal death and discuss what can be done to prevent it.
