Anaphylaxis is an immediate-type allergic reaction involving the whole organism. It is the most life-threatening allergic condition. Although there are few exact epidemiological data regarding prevalence, estimates regarding insect sting anaphylaxis range from 1-3% in the general population, but much higher values have been reported by some authors for food and drug-induced anaphylaxis. Anaphylaxis is the main acute killer of allergic individuals. Although anaphylaxis was discovered at the beginning of the 20th century, there are still many unresolved issues. These include non-IgE-mediated anaphylactoid reactions, non-immunologically mediated anaphylactoid (pseudo-allergic) reactions, pathophysiological events at the microcirculatory level, appropriate therapy for the acute reaction, strategies for prevention, public education about the problem and new approaches to prevention and therapy at the IgE level. All these subjects are discussed in this book. Since anaphylaxis occurs acutely and is unforeseen, it is very difficult to organize controlled studies regarding therapy and prevention. Over the last several years, immunologists have re-discovered the importance of regulatory lymphocytes, formerly termed 'suppressor cells'. Many recent reports have documented their existence, effector functions and potential therapeutic benefits in autoimmunity and transplantation. However, even though modern techniques have allowed us to get a much more detailed picture of these cells, they are still highly controversial. Several unresolved issues responsible for this dilemma are discussed in this book: it is difficult to grow and clone such cells, their phenotypes and effector functions are diverse and can sometimes easily be lost, and it is not well understood how they interact with antigen-presenting cells. This book contains contributions from leading investigators from around the world, including lively discussion of the current state of the art in studies of regulatory lymphocytes. Topics featured are the physiological control of autoimmunity, the role of antigen-specific cells in various diseases and disease models and effector mechanisms. Generation and Effector Functions of Regulatory Lymphocytes

Environmental statistics is a rapidly growing discipline that is important not only as a division of professional and academic statistics, but also in the accumulation of data on environmental effects and in the formulation of environmental policy by government agencies. This book features contributions from policy makers, scientists actively involved in collection of data, and professional, academic and government statisticians. These explore the interfaces between the different areas of application of environmental statistics and consider the future applications of methods arising from this discipline. Some of the chapters deal specifically with environmental models and methods. Here, the contributors explain specialized statistical techniques that have been developed for analysing environmental data and mathematical models that have been applied to the problem of integrating large amounts of environmental data into a coherent picture of particular environmental systems. Environmental Statistics

The potential lipotoxic effect of accumulation of fatty acids in non-adipose tissues is thought to be a major component in the development of insulin resistance. Chronic exposure to high concentrations of free fatty acids in the blood affects pancreatic β cell function, insulin secretion and lipid synthesis in the liver, and storage in adipose tissue. Maintaining the normal levels of fatty acids requires coordinated regulation between the liver, adipose tissue and skeletal muscle. This book deals with the molecular aspects of fatty acid action in obesity and insulin resistance. The topics include lipid metabolism and adipose tissue biology, and β cell function and insulin resistance. Chapters deal with the molecular genetics and molecular physiology of energy homeostasis.

Fatty Acid and Lipotoxicity in Obesity and Diabetes

This book explores existing and potential strategies for using the genome sequences of human, mouse, other vertebrates and human pathogens to solve key problems in the treatment of immunological diseases and chronic infections. The assembled genome sequences now provide important opportunities for solving these problems, but a major bottleneck is the identification of key sequences and circuits controlling the relevant immune reactions. This will require innovative, interdisciplinary and collaborative strategies of a scale and complexity we are only now beginning to comprehend. The Nature and Origin of Amyloid Fibrils

Ciba Foundation Symposium 123 Antidepressants and Receptor Function Chairman: Dennis Murphy, 1986 Depression is a common and often debilitating affective disorder. Attempts to develop effective antidepressants have a long history, but many questions remain about the mechanisms of action of such treatments and about the aetiology and pathophysiology of depression itself. Early observations centred attention on central monoamine systems, and animal studies suggested that changes in beta-adrenoceptor responsiveness were a common effect of antidepressant therapies. More recent research has encompassed many different central and peripheral receptors, time-dependent adaptational events at synapses, and the functional significance of changes in neurotransmitter systems in both humans and experimental animals. Such pharmacological studies aimed ultimately at elucidating the neurochemical basis of depression and of promoting new therapeutic approaches, provide the focus of this symposium volume. Many different methods of investigating the links between monoamine systems, depression and antidepressant treatments are described. Recent studies of receptors and of monoamine uptake sites in the brain and the periphery (e.g. in platelets and fibroblasts) are reviewed, with emphasis on alpha and beta adrenoceptors, [3H]imipramine-binding sites and serotonin receptors. The results of monitoring amine metabolites in cerebrospinal fluid and of measuring neuroendocrine, physiological and behavioural responses to pharmacological challenge are presented, providing information on monoaminergic function in depressed patients and experimental animals before, during and after treatment with antidepressant drugs or electroconvulsive shock. Genetic influences on receptor density are also discussed, as is the relevance to human depressive illness of animal models, including stress-induced behavioural depression in rats and responses to social stressors in rhesus monkeys. This book should be of interest to neuropharmacologists, psychopharmacologists, clinical pharmacologists, behavioural scientists, psychiatrists and neuroscientists.

Antidepressants and Receptor Function

Sensory Guidance of Movement Chairman: Mitchell Glickstein, 1998 In the past few years there has been an increasing recognition of the multiplicity of sensory and motor areas of the cerebral cortex. However, still relatively little is known about the way in which sensory areas are functionally linked to motor areas. On the basis of current anatomical evidence, there are three major pathways involved in this linking. One of these routes is by way of cortico-cortical links, beginning in the primary sensory areas of the cortex, and connecting via a series of synaptic relays to motor or premotor areas. There are also two massive subcortical routes. One of these involves the basal ganglia, the other the cerebellum. This book focuses on current research on the structure and functions of these three pathways and their role in the sensory guidance of movement. Motor psychophysicists have made progress in characterizing the nature of movements such as reaching and grasping, and how such movements are modified by incoming sensory information. Anatomical studies have revealed important new information about the ways in which sensory information is relayed to the basal ganglia and cerebellum. There is now a volume of scanning evidence about the activity of brain areas in humans and recordings from individual neurons in animals during sensory guided movement. This book summarizes much of this recent knowledge and provides a forum for suggesting new avenues for further study. The topics covered also have important implications for understanding the role of these pathways in human disease.

Sensory Guidance of Movement

This title provides an extremely helpful analysis of genes that may be associated with autoimmunity, and answers questions such as how these genes can be identified, and how the functions of the gene products can be elucidated. Incorporating data on disease-associated chromosomal loci that has been accumulated from inbred mice, the title: descibes how some susceptibility loci may be common to many diseases, whereas others are relatively disease specific discusses the importance of developing criteria for establishing the significance of these different categories of disease-associated loci.

Genetics of Autoimmunity

Parkinson's disease, Alzheimer's disease, and motor neuron disease share a significant common feature: selective death of neurons in restricted regions of the brain. This international symposium, held by the Ciba Foundation in 1986, is the first to bring together neurophysiologists working on neuronal death and neuropathologists dealing with human degenerative brain disease. Participants describe the causes and sequence of events leading to neuronal death and discuss what can be done to prevent it. Among the topics covered are recent advances in the understanding of agents such as trophic factors, excitotoxins and poisons that are known to be involved in neuronal death; examples of neuronal death during normal development; and the role played by endocrine mechanisms and neuronal activity. Also considers trophic factors controlling the survival of neuronal transplants and the therapeutic prospects for tissue transplantation.
