Ciba Foundation Symposium 123 Antidepressants and Receptor Function Chairman: Dennis Murphy, 1986 Depression is a common and often debilitating affective disorder. Attempts to develop effective antidepressants have a long history, but many questions remain about the mechanisms of action of such treatments and about the aetiology and pathophysiology of depression itself. Early observations centred attention on central monoamine systems, and animal studies suggested that changes in beta-adrenoceptor responsiveness were a common effect of antidepressant therapies. More recent research has encompassed many different central and peripheral receptors, time-dependent adaptational events at synapses, and the functional significance of changes in neurotransmitter systems in both humans and experimental animals. Such pharmacological studies aimed ultimately at elucidating the neurochemical basis of depression and of promoting new therapeutic approaches, provide the focus of this symposium volume. Many different methods of investigating the links between monoamine systems, depression and antidepressant treatments are described. Recent studies of receptors and of monoamine uptake sites in the brain and the periphery (e.g. in platelets and fibroblasts) are reviewed, with emphasis on alpha and beta adrenoceptors, [3H]imipramine-binding sites and serotonin receptors. The results of monitoring amine metabolites in cerebrospinal fluid and of measuring neuroendocrine, physiological and behavioural responses to pharmacological challenge are presented, providing information on monoaminergic function in depressed patients and experimental animals before, during and after treatment with antidepressant drugs or electroconvulsive shock. Genetic influences on receptor density are also discussed, as is the relevance to human depressive illness of animal models, including stress-induced behavioural depression in rats and responses to social stressors in rhesus monkeys. Antidepressants and Receptor Function

Sensory Guidance of Movement Chairman: Mitchell Glickstein, 1998 In the past few years there has been an increasing recognition of the multiplicity of sensory and motor areas of the cerebral cortex. However, still relatively little is known about the way in which sensory areas are functionally linked to motor areas. On the basis of current anatomical evidence, there are three major pathways involved in this linking. One of these routes is by way of cortico-cortical links, beginning in the primary sensory areas of the cortex, and connecting via a series of synaptic relays to motor or premotor areas. There are also two massive subcortical routes. One of these involves the basal ganglia, the other the cerebellum. This book focuses on current research on the structure and functions of these three pathways and their role in the sensory guidance of movement. Motor psychophysicists have made progress in characterizing the nature of movements such as reaching and grasping, and how such movements are modified by incoming sensory information. Anatomical studies have revealed important new information about the ways in which sensory information is relayed to the basal ganglia and cerebellum. There is now a volume of scanning evidence about the activity of brain areas in humans and recordings from individual neurons in animals during sensory guided movement. This book summarizes much of this recent knowledge and provides a forum for suggesting new avenues for further study. The topics covered also have important implications for understanding the role of these pathways in human disease.

Sensory Guidance of Movement

Incontinence is a very common and often devastating problem, but one that goes largely unacknowledged. In order to elucidate the underlying mechanisms of this major clinical condition, this symposium brought together neuroscientists working on the basic biology of the bladder and bowel and clinicians dealing with the various manifestations of urinary and fecal incontinence. Higher-Order Processing in the Visual System

The number of factors implicated in the regulation of cell proliferation and differentiation is already considerable and more are continually being identified. This book concentrates on tumor necrosis factor (cachectin) and lymphotoxin, but includes observations of their interactions with other cytokines, especially the interferons and interleukins. TNF can be either cytostatic or cytotoxic to cultured cell lines, and a variety of mechanisms are proposed, ranging from DNA fragmentation to activation of phospholiopases. TNF is also reported to stimulate the growth of normal fibroblasts in vivo. The in vivo actions of TNF described here include haemorrhagic necrosis of tumors, cachexia, effects on the vasculature, and antiparasitic and antiviral activities. Cigarette smoking is estimated to lead to 4.9 million premature deaths per year worldwide. This is predicted to rise to 10 million by 2020. In western countries it kills half of all smokers who fail to stop. The prevailing model for tobacco addiction is that nicotine from cigarettes rewards smoking and punishes abstinence, tapping into a motivational system of operant conditioning that requires no conscious awareness. However, there are also accounts which involve cognitive biases and the effect of nicotine on impulse control. The brain pathways involved have been studied extensively, but the role of different nicotine receptor subtypes and other neurotransmitter systems is still subject to debate. In western countries, cigarette smoking as an adult has a heritability of 30-50% and candidate genes have been identified that may contribute in part to addiction susceptibility. Many socio-cultural correlates of cigarette smoking have been established, but a comprehensive model that accounts for these and links them with the psychobiological aspects of nicotine addiction has not been forthcoming. Structured behavioural support programmes aid cessation attempts, as do a number of pharmacotherapies, most notably nicotine replacement treatments and bupropion, but the underlying mechanisms are unclear. This book deals with the problems involved in understanding and treating nicotine and tobacco addiction. Topics covered include the nature of the worldwide health problem posed by cigarette smoking, the psychodynamics of cigarette addiction, and the basic pharmacology and biochemistry of nicotine and its effect on the brain. Further chapters analyse the genetic basis of susceptibility to nicotine addiction. Finally, the contributors address approaches to therapy. A continuing theme in the discussions throughout the book is how best to treat nicotine addiction, given that many smokers would like to stop smoking but are unable to do so because of their addiction. This book will be of great value to all psychologists and psychiatrists working on addiction, specifically to nicotine but also to other compounds and behaviours. Understanding Nicotine and Tobacco Addiction

Over the last several years, immunologists have re-discovered the importance of regulatory lymphocytes, formerly termed 'suppressor cells'. Many recent reports have documented their existence, effector functions and potential therapeutic benefits in autoimmunity and transplantation. However, even though modern techniques have allowed us to get a much more detailed picture of these cells, they are still highly controversial. Several unresolved issues responsible for this dilemma are discussed in this book: it is difficult to grow and clone such cells, their phenotypes and effector functions are diverse and can sometimes easily be lost, and it is not well understood how they interact with antigen-presenting cells. Anaphylaxis is an immediate-type allergic reaction involving the whole organism. It is the most life-threatening allergic condition. Although there are few exact epidemiological data regarding prevalence, estimates regarding insect sting anaphylaxis range from 1-3% in the general population, but much higher values have been reported by some authors for food and drug-induced anaphylaxis. Anaphylaxis is the main acute killer of allergic individuals. Although anaphylaxis was discovered at the beginning of the 20th century, there are still many unresolved issues. 'Homology' as a concept became increasingly elusive during the course of the 20th century. The central debates and controversies concern both fundamental definitions and the nature of the criteria by which homology is judged. Attempts to move away from comparative morphology to ideas based on developmental pathways have tended to founder on the fact that developmental pathways evolve and that similar cells or tissues or structures in animals will often have different developmental origins. Environmental statistics is a rapidly growing discipline that is important not only as a division of professional and academic statistics, but also in the accumulation of data on environmental effects and in the formulation of environmental policy by government agencies. These explore the interfaces between the different areas of application of environmental statistics and consider the future applications of methods arising from this discipline.
